Conclusion: Our findings show that ketamine and esketamine may be more efficacious than placebo at 24 hours. How these findings translate into clinical practice, however, is not entirely clear. The evidence for use of the remaining glutamate receptor modulators is limited as very few trials were included in the meta-analyses for each comparison and the majority of comparisons included only one study. Long term non-inferiority RCTs comparing repeated ketamine and esketamine, and rigorous real-world monitoring are needed to establish comprehensive data on safety and efficacy.

What got worse for some people
In their own words
“long courses of Sarcosine (daily for a month+ at a time) helped tremendously and permanently, however when I tried it again more recently it triggered anxiety and overthinking.”
“just 1 dose of sarcosine had me feeling incredible. It felt as though I could breathe again. Anxiety lifted, mood was relaxed but NOT tired”
“I've only been on Sarcosine (1 gram three times a day) for almost a week now, but it seems to have completely obliterated my old, self-destructively negative thinking patterns”
